تأثیر تمرین تناوبی پر شدت بر بیان ژن های پرو و آنتی آپوپتیک در بافت قلبی موش های نر پیر نژاد C57BL/6

چکیده

هدف: ژنهای Bax و Bcl-2 ازجمله ژن های مهم مسیر داخلی آپوپتوز می باشند. از طرفی، تأثیر فعالیت های ورزشی تناوبی و پر
شدت روی سازوکارهای دخیل در فرآیند پیری کمتر موردتوجه قرارگرفته اند. لذا، پژوهش حاضر باهدف تعیین تأثیر تمرین تناوبی
پر شدت بر بیان ژن های پرو و آنتی آپوپتیک ) Bax و Bcl2 ( در بافت قلبی موش های نر پیر انجام شد.
روش ها: این مطالعه تجربی روی 20 سر موش نر 24 ماهه نژاد C57BL/6 انجام شد. حیوانات به طور تصادفی به دو گروه کنترل
( 10 سر( و آزمایش ) 10 سر( تقسیم شدند. گروه آزمایش به مدت 4 هفته و هر هفته 5 جلسه به تمرین تناوبی پر شدت پرداختند.
در ابتدا تمرین تناوبی با شدتی برابر با 85 درصد سرعت بیشینه شروع شده و در ادامۀ برنامه تناوبی، هفته ای پنج درصد به سرعت
افزوده شد و دو هفته آخر با 95 درصد سرعت خاتمه یافت. 48 ساعت پس از آخرین جلسه ی تمرین، بافت قلبی جدا شد. بیان
ژن های BCL-2 و Bax بهوسیله SYBR-Green Real-time PCR موردبررسی قرار گرفت.
نتایج: نتایج حاصل از Real Time-PCR نشان داد که میانگین میزان بیان نسبی ژن پروآپوپتوتیک Bax در گروه آزمایش به طور
معنی داری کمتر از گروه کنترل بود ) 022 / 0 ± 618 / 0 واحد در مقابل 1 واحد، 05 / 0P< (. همچنین، میانگین میزان بیان ژن
آنتی آپوپتوتیک Bcl2 در گروه آزمایش، به طور معنی داری بیشتر از گروه کنترل بود ) 084 / 0 ± 824 / 1 واحد در مقابل 1 واحد،
05 / 0P< .)
نتیجه گیری: نتایج این مطالعه نشان داد که تمرین تناوبی پر شدت با تعدیل بیان ژنهای پرو و آنتی آپوپتیک ) Bax و Bcl2 ( در
کاهش مرگ برنامه ریزی شده در بافت قلبی موش های پیر مؤثر است.

Purpose: Bax and BCL-2 genes are important genes that involve in the intrinsic pathway of apoptosis. Although the effects of interval and intensive exercises on the mechanisms involved in the gaining processes are less considered. So, the aim of the present study was to investigate the effect of high-intensity interval training on the expression of pro and anti-apoptotic genes (Bax and BCL-2) in myocard in aged male mice.
Methods: This experimental study was performed on twenty 24- months old C57BL/6 mice. Animals were randomly assigned to the control (n=10) and experimental groups (n=10). The experimental group performed high interval intensity training for four weeks (five sessions per week). At first, the interval training started at the intensity equal to 85 percent of maximum speed and in the continuation of interval training, 5 percent was added weekly and the last two weeks ended with 95 percent rate. Forty-eight hours after the last training session, myocardial muscle was removed. Bax and BCL-2 genes expression were investigated by SYBR-Green Real-time PCR analysis.
Results: The results of Real Time-PCR showed that the average relative expression of pro-apoptotic Bax gene was significantly lower in the experimental group than in the control group (0.618±0.022 vs. 1 unit; P<0.05). Also, the average relative expression of Anti-apoptotic Bcl2 gene expression was higher in the experimental group than in the control group (1.824±0.084 vs. 1 unit; P< 0.05).
Conclusion: The results showed that high-intensity interval training by modifying the expression of pro and anti-apoptotic genes (Bax and BCL-2) may be effective in reducing apoptosis in cardiac tissue of aged mice.
