تاثیر تمرین هوازی و هوای آلوده به ذرات کربن سیاه بر بیان ژنهای κB-NF و α-TNF بافت هیپوکامپ مغز موشهای صحرایی نر

چکیده

هدف:هدف از این پژوهش بررسی تاثیر تمرین هوازی و هوای آلوده به ذرات کربن سیاه بر بیان ژنهای κB-NF و α-TNF هیپوکامپ
مغز موشهای صحرایی نر بود.
روشها: بیست و چهار موش صحرایی نر ویستار )با میانگین توده بدنی 97/26±29/279 گرم( بهطور تصادفی در چهار گروه: الف-
بدون تمرین و بدون قرار گرفتن در معرض هوای آلوده به ذرات کربن سیاه؛ ب- تمرین هوازی؛ ج- قرار گرفتن در معرض ذرات؛ و
د- تمرین و با قرار گرفتن در معرض ذرات کربن سیاه قرار گرفتند. برای قرار دادن حیوانات در معرض ذرات کربن سیاه با قطر
کمتر از 10 میکروناز دستگاه تزریق ذرات و اتاقک استفاده شد. مدت تمرین هوازی با 50 درصد سرعت بیشینه هر گروه 4 هفته
بود و 24 ساعت پس از آخرین جلسه تمرینی و قرارگیری در معرض ذرات کربن سیاه حیوانات قربانی شدند. با استفاده از روش
Pcr-time Real بیان ژن κB-NF و α-TNF در بافت هیپوکامپ موشها ارزیابی شد. برای تعیین معنادار بودن تفاوت بین گروهها
از تحلیل واریانس دوطرفه و آزمون تعقیبی LSD استفاده شد.
نتایج: تفاوت معناداری بین گروههای الف و ب )001/0=P ،)الف و د )003/0=P )در مقدار α-TNF وجود داشت. تفاوت معناداری
نیز بین گروههای الف و ب )008/0=P )در مقدار κB-NF دیده شد.
نتیجهگیری: چهار هفته تمرین هوازی و هوای آلوده به ذرات کربن سیاه باعث تعدیل عوامل التهابی κB-NF و α-TNF میشود. به
نظر میرسد این تغییرات در بخشی با کاهش خطر ابتال به بیماریهای عصبی مغزی همراه باشد.

چکیده [English]

Purpose: The purpose of this study was to investigate the effect of aerobic exercise and black carbon particles
on gene expression of NF-κB and TNF-α in rats.
Methods: Twenty-four male Wistar rats with an average body mass 279.29±26.97 g were randomly divided
into 4 groups A: without exposure PM10 and aerobic exercise, B: aerobic training, C: exposure to PM10
carbon black (5 mg/m3; per rat), D: and aerobic exercise concomitantly with exposure to PM10 carbon black.
Twenty-four hours after the last training session and exposure, the animals were sacrificed. Real timePcr technique was used. Two-way ANOVA and post hoc LSD was used to determine the significance of
differences between groups.
Results: There was significant difference between groups A and B (P=0.001) and B and D (P=0.003) in the
amount of TNF-α. Significant difference between A and B groups (P=0.008) in the amount of NF-κB was
observed.
Conclusions: four weeks of aerobic exercise and PM10 exposure lead to modify the inflammatory factors of
NF-κB and TNF-α. It seems that these changes lead to reducing the risk of neurodegenerative diseases.
