اثر شش هفته تمرین مقاومتی با مکمل‌یاری پروتئین وی بر اسفنگوزین- 1- فسفات و هایپرتروفی ماهیچة دوقلوی موش‌های صحرایی نر جوان نژاد ویستار

چکیده
سابقه و هدف: امروزه افزایش حجم ماهیچه و اجرای روش‌هایی جهت بهبود بازیافت پس از تمرین از اهمیت ویژه‌ای برخوردار است. تمرین مقاومتی نقش ویژه‌ای در افزایش حجم ماهیچه و هایپرتروفی دارد. دریافت انواع مختلفی از مکمل‌ها، همچون پروتئین وی پس از تمرین مقاومتی نیز می‌تواند به این عمل کمک کند. اما از بین عوامل مختلف، اسفنگوزین مونو فسفات (S1P) نقش ساختاری در تنظیم، تمایز سلول و هایپرتروفی دارد و این عامل می‌تواند با تمرین و دریافت مکمل نقش مهم و ارزنده‌ای در هایپرتروفی داشته باشد. پروتئین وی نیز در ترکیب با تمرین مقاومتی می‌تواند پروتئین‌سازی ماهیچه، بهبود تمرین و بازیافت در افراد جوان‌تر را افزایش دهد. از این‌رو، هدف این پژوهش بررسی تأثیر شش هفته تمرین مقاومتی با مکمل‌یاری پروتئین Whey بر S1P و هایپرتروفی ماهیچة دوقلوی موش‌های صحرایی نر جوان نژاد ویستار بود.
مواد و روش‌ها: در این پژوهش تجربی 23 سر موش‌ صحرایی هشت‌هفته‌ای با دامنة وزن 250 تا 270 گرم به‌صورت تصادفی در چهار گروه تمرین- مکمل، تمرین مقاومتی، شم و کنترل جایگزین شدند و به مدت شش هفته با روش تمرینی پنج‌جلسه‌ای در سه نوبت و پنج تکرار به شکل بالا رفتن از نردبان یک‌متری و حمل وزنة متصل به دم تمرین داده شدند. در گروه تمرین- مکمل محلول مکمل پروتئین وی نیز گاواژ شد. بیان ژن S1P به روش Real Time-PCR و هایپرتروفی ماهیچه با ترازوی مخصوص اندازه‌گیری شد. در تحلیل داده‌ها از آزمون شاپیروویلک، واریانس یکراهه و تعقیبی شفه در سطح 05/0 P≤ استفاده شد.
نتایج: نتایج نشان داد که بیان ژن S1P (001/0=p)، حجم ماهیچه (016/0=p و 1-RM (001/0=p) درگروه تمرین- مکمل نسبت به گروه تمرین و شم به‌طور معناداری افزایش یافت. با این حال، تفاوت معناداری در میزان حجم ماهیچه در بین گروه‌های تمرین و شم با گروه کنترل مشاهده نشد (6/0P=).
نتیجه‌گیری: به‌طور کلی، نتایج این تحقیق نشان می‌دهد که ترکیب تمرین با مصرف مکمل پروتئین وی ممکن است از طریق تأثیر بر بیان ژن S1P در بهبود برخی سازگاری‌های عضلانی نقش داشته باشد. با وجود این، به‌دلیل محدودیت‌های تحقیق، از جمله امکان محدود بودن حجم نمونه و شرایط اجرای پژوهش، تعمیم این نتایج به سایر جمعیت‌ها نیازمند احتیاط است. انجام پژوهش‌های آینده با حجم نمونة بیشتر، دوره‌های مداخلة طولانی‌تر و بررسی سازوکار‌های مولکولی مرتبط می‌تواند به تبیین دقیق‌تر این یافته‌ها کمک کند.

The effect of six weeks of resistance training with whey protein supplementation on sphingosine 1-phosphate and gastrocnemius muscle hypertrophy in young male Wistar rats

Background and Purpose: Currently, increasing muscle mass and implementing methods to improve recovery after training are of particular importance. Resistance training plays a special role in increasing muscle mass and hypertrophy. Taking various types of supplements, such as whey protein after resistance training, can also help in this process. Sphingosine monophosphate (S1P) has a structural role in regulation, cell differentiation and hypertrophy, and this factor can play an important role in hypertrophy with training and supplementation. Whey protein, in combination with resistance training, can increase muscle protein synthesis, improve training and recovery in younger individuals. Therefore, this study aimed to investigate the combined effect of six weeks of resistance training with whey protein supplementation on S1P and hypertrophy of the gastrocnemius muscle of young male Wistar rats.
Materials and Methods: In this experimental study, 23 eight-week-old rats with a weight range of 250 to 270 grams were randomly assigned to four groups: supplement plus training, resistance training, sham, and control. They were trained for six weeks, five sessions per week. Each session included performance of 3 sets of 5 repetitions, climbing a one-meter ladder while carrying a weight attached to the tail. Whey protein supplement solution was administered by gavage in the supplementa plus training group. S1P gene expression was measured by Real Time-PCR and muscle hypertrophy was measured through weighing the gastrocnemius muscle. Data analysis were performed by using one-way variance, and Scheffe post-hoc test.
Results: Data analysis revealed a significant increase in S1P gene expression (P=0.001), muscle volume (P=0.016), and 1-RM (P=0.001) in training-supplement group compared to both training and sham groups. However, no significant differences in muscle volume were detected between the training and sham groups in comparison to the control group (p=0.6).
Conclusions: Overall, the findings of this study suggest that combining exercise with supplementation might induce muscle adaptations through its effects on S1P gene expression. However, due to study limitations, caution is warranted in generalizing these results to the population. Future studies with larger sample sizes, longer intervention periods, and concentrating on molecular mechanisms could help to shed some more lights on mechanisms.
