English

Background and Purpose: Obesity, as a chronic metabolic disorder, is associated with the activation of inflammatory pathways, impaired energy regulation, and increased production of reactive oxygen species. Obesity-induced inflammation and exposure to acrylamide may disturb the metabolic rate of liver tissue. In this study, the effect of the synergistic combination of enriched medium and cineole on the expression of key genes related to inflammation, metabolism, and oxidative stress were evaluated in an obese animal model induced by a high-fat diet containing acrylamide.

Materials and Methods: 54 C57BL/6J mice were subjected to different treatments for eight weeks. Mice were randomly assigned to nine equal groups (n, 6 per group): (1) healthy control; (2) obese mice fed a high-fat diet containing acrylamide (45% high-fat diet heated and enriched with 1000 ppm acrylamide); (3) obese mice fed a high-fat diet without acrylamide (45% high-fat diet); (4) obese mice fed acrylamide-containing-diet and undergoing voluntary physical activity in an enriched environment (8 weeks, 5 days/week, 60-90 minutes/day, at low to moderate intensity); (5) obese mice fed an acrylamide-containing diet and treated with cineole (10 μM concentration by gavage); (6) obese mice, undergoing voluntary physical activity in an enriched environment; (7) obese mice treated with cineole; (8) obese mice fed an acrylamide-containing diet, undergoing voluntary physical activity in an enriched environment, and receiving cineole; (9) obese mice exposed to an enriched environment and treated with cineole without acrylamide exposure. Drug affinity analyses were based on pharmacophore modeling and molecular binding. Advanced biological data analysis was performed using microarray datasets. Gene expression was measured by Real-Time PCR following RNA extraction from liver tissue.

Results: voluntary physical activity in enriched environment complained with cineole significantly increased the expression (P
